Comprehensive Evaluation of Lilium Pollen Viability and Germination Methods

Abstract

The assessment of Lilium pollen viability and germination is critical for effective breeding and cultivation practices. In this study, three varieties of lily were utilized, namely L. oriental 'Acapulco', L. longiflorum 'Woori Tower', and L. martagon 'Arabian Knight'. The pollen was collected during the flowering period, carefully wrapped in parchment paper, and stored in tubes filled with silica gel at a temperature of -20 degrees Celsius for a year. Two techniques were employed to evaluate the pollen viability and germination: the Alexander stain method and pollen germination media. The results showed that most of the tested varieties exhibited high fertility, indicating robust pollen viability, after applying the Alexander stain. However, during the pollen germination media test, only L. martagon demonstrated significant pollen tube elongation, suggesting that it has particular germination capabilities. These findings highlight the importance of employing multiple assessment techniques to comprehensively evaluate Lilium pollen viability and germination potential, which can provide valuable insights for breeders and horticulturists to improve Lilium cultivation and breeding strategies. It is important to note that the long-term storage of pollen plants may produce different experimental results depending on the storage method and pollen media conditions. Therefore, it is essential to develop a suitable long-term storage method for pollen plants and identify the optimal potting medium conditions for lily pollen.
